    Studies of a weak polyampholyte at the air-buffer interface: The effect of varying pH and ionic strength

    We have carried out experiments to probe the static and dynamic interfacial properties of Ī²\beta--casein monolayers spread at the air-buffer interface, and analysed these results in the context of models of weak polyampholytes. Measurements have been made systematically over a wide range of ionic strength and pH. In the semi-dilute regime of surface concentration a scaling exponent, which can be linked to the degree of chain swelling, is found. This shows that at pH close to the isoelectric point, the protein is compact. At pH away from the isoelectric pH the protein is extended. The transition between compact and extended states is continuous. As a function of increasing ionic strength, we observe swelling of the protein at the isoelectric pH but contraction of the protein at pH values away from it. These behaviours are typical of a those predicted theoretically for a weak polyampholyte. Dilational moduli measurements, made as a function of surface concentration exhibit maxima that are linked to the collapse of hydrophilic regions of the protein into the subphase. Based on this data we present a configuration map of the protein configuration in the monolayer. These findings are supported by strain (surface pressure) relaxation measurements and surface quasi-elastic light scattering (SQELS) measurements which suggest the existence of loops and tails in the subphase at higher surface concentrations.Comment: Submitted to J.     An investigation of conducted and radiated emissions from a hollow-cathode plasma contactor

    An investigation conducted on the electrical interference induced by the operation of a hollow-cathode plasma contractor in a ground-based facility is described. The types of electrical interference, or noise, which are important to Space Station Freedom designers are classified as either conducted or radiated emissions. The procedures required to perform conducted and radiated emission measurements on a plasma contactor are examined. The experimental data obtained are typically examined in the frequency domain (i.e. amplitudes of the noise fluctuations versus frequency). Results presented indicate the conducted emissions, which are the current fluctuations from the contactor into the space station wiring, are affected by operating parameters such as expellant flow rate and discharge current. The radiated emissions, which are the electromagnetic waves induced and emitted by the contactor, appear to be influenced by the contactor emission current. Other experimental results suggest possible sources which are responsible for the observed noise. For example, the influence of the plasma environment downstream from the contactor on noise emission levels is described. In addition, a brief discussion is given on the correlation between conducted and radiated emissions and the mechanisms through which both are influenced by the plasma downstream of the contactor

The reduction in available surface area as the gas flows toward the center of the disk reduces the oxygen removal area proportionally to provide for a more uniform removal of oxygen

    Multi-dimensional classification of biomedical text: Toward automated, practical provision of high-utility text to diverse users

    Motivation: Much current research in biomedical text mining is concerned with serving biologists by extracting certain information from scientific text. We note that there is no ā€˜average biologistā€™ client; different users have distinct needs. For instance, as noted in past evaluation efforts (BioCreative, TREC, KDD) database curators are often interested in sentences showing experimental evidence and methods. Conversely, lab scientists searching for known information about a protein may seek facts, typically stated with high confidence. Text-mining systems can target specific end-users and become more effective, if the system can first identify text regions rich in the type of scientific content that is of interest to the user, retrieve documents that have many such regions, and focus on fact extraction from these regions. Here, we study the ability to characterize and classify such text automatically. We have recently introduced a multi-dimensional categorization and annotation scheme, developed to be applicable to a wide variety of biomedical documents and scientific statements, while intended to support specific biomedical retrieval and extraction tasks

    STATISTICAL ISSUES IN THE ANALYSIS OF MICROBIAL COMMUNITIES IN SOIL

    Corn and soybean production dominates the agricultural systems of the mid-western United States. Studies have found that when a single crop species is grown continually, without the rotation of other crops, yield decline occurs. At present, this phenomenon, remains poorly understood, but there are possible links to microbial community dynamics in the associated rhizosphere soil. In this study, corn plants were grown in disturbed and undisturbed soils with a 24 year history of growth as a mono culture crop or two crops grown in annual rotation. Characteristic profiles of the microbial communities were obtained by denaturing gradient gel electrophoresis of polymerase chain reaction amplified 16S rDNA from soil extracted DNA. This problem is approached as the statistical analysis of high-dimensional multivariate binary data with an emphasis on modeling and variable selection

    Effect of chloride lons on the reaction of diphenylamine with DNA

    The sensitivity of the Dische test (diphenylamine) for DNA is reduced by the presence of chloride ions, which cause formation of a new reaction product whose absorption maximum is at lower wavelength, and also a reduction in the total chromogenic potential.     <p>Abstract</p> <p>Background</p> <p>We present a probabilistic topic-based model for content similarity called <it>pmra </it>that underlies the related article search feature in PubMed. Whether or not a document is about a particular topic is computed from term frequencies, modeled as Poisson distributions. Unlike previous probabilistic retrieval models, we do not attempt to estimate relevanceā€“but rather our focus is "relatedness", the probability that a user would want to examine a particular document given known interest in another. We also describe a novel technique for estimating parameters that does not require human relevance judgments; instead, the process is based on the existence of MeSH <sup>Ā® </sup>in MEDLINE <sup>Ā®</sup>.</p> <p>Results</p> <p>The <it>pmra </it>retrieval model was compared against <it>bm25</it>, a competitive probabilistic model that shares theoretical similarities. Experiments using the test collection from the TREC 2005 genomics track shows a small but statistically significant improvement of <it>pmra </it>over <it>bm25 </it>in terms of precision.</p> <p>Conclusion</p> <p>Our experiments suggest that the <it>pmra </it>model provides an effective ranking algorithm for related article search.</p
